Lipid nanoparticles can be tough to transport
Lipids, typically in the form of lipid nanoparticles (LNPs), are a key component of the delivery systems for messenger RNA (mRNA) vaccines. This includes recently approved vaccines from Pfizer and Moderna currently being used in the fight against COVID-19.
LNPs deliver COVID-19’s spike protein mRNA directly to our cells, which then replicate the protein and cause our bodies to initiate an immune response to the virus. This helps protect us if we are infected with COVID-19.
Lipids, like glycerides, cholesterols and proteins, easily stick to surfaces (adsorb) when in a solution. This can be a serious problem that only worsens as the concentration of the lipid solution increases or as the particle size decreases. Lipids also have a long history of sticking to the insides of bottles and other storage containers, reducing yield when the lipids are emptied out.
Recent developments in LNP manufacturing now require the lipid to be dissolved in an organic solvent (typically 90% ethanol). However, using these solvents increases the risk of contaminants being leached from commonly used polymer containers.
